  • This song finds The Lonely Island addressing hip-hop's machismo, which often comes out as homophobia. Andy Samberg explained to Spinner UK: "'No homo' has become so ubiquitous a phrase that we felt like it had to be addressed. We saw little kids saying it, like, 'Wanna come over and play, no homo' and it just seemed ridiculous so we wrote the song. We hope the reaction to it is laughter. That's always the desired reaction."
  • The Lonely Island - the team of Andy Samberg, Akiva Schaffer, and Jorma Taccone - were still working for Saturday Night Live when they released "No Homo" on their second album, Turtleneck & Chain. They joined the show as a team in 2005, Samberg as a cast member, Schaffer and Taccone as writers. Many of their songs were written for sketches, but this one didn't have a video to go with it and wasn't aired on SNL.
  • The riff is sampled from "I'm Not To Blame," a 1970 track by Bobby Byrd produced by James Brown.
